Disclaimer: I'm an AI trainer. It depends what you're asking it. The more quantitative your prompt (e.g., "solve this system of equations:" is entirely quantitative), the less a requested persona will affect the substance of the prompt.
Pure maths/science problems as prompts, with a requested persona, will elicit responses that have token, tacked-on characterisation in the introduction and conclusion and a completely unaffected middle section which will be the reasoning and solution.
The more qualitative the prompt (e.g., "write a sonnet" is almost entirely qualitative), the more a persona may matter, but a requested persona should never affect the core of the response very much.
